Reconstitution and storage of lyophilized research peptides

Lyophilized material is stable in transit and sensitive after it is opened. Correct handling in the laboratory preserves the purity the certificate reports.

Peptides are supplied as a lyophilized (freeze-dried) cake precisely because the dry state is stable. The moment a vial is reconstituted, its clock starts. The practices below describe laboratory handling of research materials and exist to keep the compound consistent with the analysis on its certificate.

On receipt

Lyophilized material ships ambient and tolerates transit. On arrival, store the sealed vial refrigerated at 2–8°C for near-term use, or at −20°C for longer holding, protected from light. Allow a cold vial to reach room temperature before opening to avoid drawing moisture into the powder.

Reconstitution

Reconstitution is performed with an appropriate sterile diluent selected for the protocol. Direct the stream against the wall of the vial rather than onto the cake, and let the material dissolve without vigorous agitation. Peptides are mechanically fragile; shaking and foaming can degrade the sequence and are avoided in favor of gentle swirling.

Concentration and diluent are determined by the researcher’s protocol. The objective of good technique is simple: dissolve the material fully without introducing shear, contamination, or moisture that the certificate did not account for.

After reconstitution

Reconstituted material is kept refrigerated and used within the working window established by the protocol. Repeated freeze-thaw cycles are minimized; where a solution must be held, single-use aliquots reduce the number of times any one fraction is warmed and re-cooled. Label aliquots with the lot number so that every working solution remains traceable to its certificate.

Why handling protects the number on the certificate

Purity, net peptide content and endotoxin results describe the material as it was released. Heat, light, moisture, mechanical stress and contamination are the variables that move a real sample away from that description. Storage and reconstitution discipline is how a laboratory keeps the vial it is using the same as the vial that was tested.
